Intended use

This ELISpot Path kit is intended for the enumeration of cells secreting human IFN-γ in response to Dengue virus peptides. For research use only. Not for use in diagnostic procedures.

Peptide pool composition

PepPool: DENV (CD4), human contains 90 defined synthetic peptides derived from the Dengue virus. Antigens represented in the peptide pool are the capsid protein (C), precursor membrane protein (prM), envelope protein (E), and the non structural (NS) proteins NS1, NS2a, NS2b, NS3, NS4a, NS4b, and NS5. The peptide pool has been designed to have a good coverage of Dengue virus serotypes 1, 2, 3, and 4, and with good HLA class II coverage.

PepPool: DENV (CD8), human contains 109 defined synthetic peptides derived from the Dengue virus. Antigens represented in the peptide pool are the capsid protein (C), envelope protein (E), and the non structural (NS) proteins NS1, NS2a, NS2b, NS3, NS4a, NS4b, and NS5. The peptide pool has been designed to have a good coverage of Dengue virus serotypes 1, 2, 3, and 4, and with good population coverage of all major HLA class I supertypes.

PBMC (200,000 cells/well) from individuals with confirmed DENV-1 or DENV-3 infection were incubated for 18 hours in the absence or precence of PepPool: DENV (CD4), human (product code 3698-1). The number of cells secreting IFN-γ was analyzed by ELISpot.

IFN-γ

Analyte description

Interferon-γ (IFN-γ) is the only type II interferon. This proinflammatory cytokine is secreted by activated T cells and NK cells. It activates macrophages and endothelial cells and regulates immune responses by affecting APCs, T cells, and B cells. Production of IFN-γ by helper T cells and cytotoxic T cells is a hallmark of the Th1-type phenotype. Thus, high-level production of IFN-γ is typically associated with effective host defense against intracellular pathogens.
